Nano-sized probes reveal how cellular structure responds to pressure

TSUKUBA, Japan, Nov 21, 2023 – (ACN Newswire) – By giving living cells a ‘nano-poke’ and monitoring the resulting changes in the intra-cellular environment, researchers have gotten their first glimpse of how whole cells respond to external mechanical pressure.

A team lead by scientists from the National Institute for Materials Science in Tsukuba, Japan, used a technique called atomic force microscopy to apply force across the surface of various cells. The method uses nanoscale probes, with tips just a few billionths of a metre in size, to measure and map how force gets distributed across the cellular surface and throughout the cell. The researchers used machine learning to analyse and model the forces they measured. They also used fixing and staining techniques to study how the force distortion affected the cell’s internal structures and the microtubules and actin filaments that make up its ‘skeleton’.

“Cells are smart materials that can adapt to various chemical and mechanical stimuli from their surroundings,” says Jun Nakanishi, one of the corresponding authors of the study and the leader of the Mechanobiology Group at the National Institute for Materials Science. That ability to adapt relies on rapid feedback mechanisms to keep the cell intact and healthy, and there’s growing evidence that the failure of this cellular response underlies a range of ailments, including diabetes, Parkinson’s disease, heart attacks, and cancer.

So far, studies of these cellular responses have been limited by the techniques used – for example, some methods require that cells be pre-fitted with sensors, so they can only measure a small part of the response. “We invented a unique way to ‘touch’ a cell with nanoscale ‘hand’, so that the force distribution over a complete cell could be mapped with nanometer resolution,” says Hongxin Wang, who is the first author of the study and JSPS postdoc in the Mechanobiology Group.

The study revealed that tensional and compressional forces are distributed across actin fibres and microtubules within the cell to keep its shape, similar to how the poles and ropes of a camping tent work. When the researchers disabled the force-bearing function of actin fibres, they found that the nucleus itself is also involved in counterbalancing external forces, highlighting the role of the internal structure of the nucleus in the cellular stress response.

The research team also compared the responses of healthy and cancerous cells. Cancer cells proved more resilient to external compression than the healthy cells, and they were less likely to activate cell death in response.

The findings not only illuminate the complex intracellular mechanics of the stress response, but the discovery of different responses in cancer cells could offer a new way to distinguish healthy and cancerous cells – a diagnostic tool based on cellular mechanics.

Hospitals currently use the size, shape, and structure of a cell in diagnosing cancer. However, these features don’t always provide enough information to tell the difference between healthy and diseased cells. “Our findings provide another way of checking cell conditions by measuring force distribution, which could dramatically improve diagnostic accuracy,” says Han Zhang, another corresponding author of the study and the senior researcher of the Electron Microscopy Group, NIMS.

TransNusa Ends 2023 as Fastest Growing Airline in South East Asia

With the launch of the Jakarta – Singapore route, TransNusa has successfully planned, developed and launched four new international routes in a short span of eight months. The airline, a low-cost carrier in its domestic market, has managed to expand its airline business to include international routes, using its own customised business model that allowed the airline to rebrand itself as a Premium Service Carrier.

TransNusa Group Chief Executive Officer, Datuk Bernard Francis, who was instrumental to developing the airline’s customised business plan and growth path, said that TransNusa has become the fastest growing airline in South East Asia due to the business plan that was developed and implemented swiftly post Covid-19.

“Post Covid-19, we initiated an intensive market research. Based on the data obtained, we knew that traveller’s behavioural pattern had changed, specifically due to the pandemic.  Our next step was to develop a customised business model for the targeted passengers. We implemented the business model on April 14th, with the launch of our first maiden international flight,” said Datuk Bernard, adding that the airline has been enjoying a significant growth pattern since April.

An aviation industry expert, who specializes in airline turnaround and revenue management, Datuk Bernard, who was part of the entourage that landed in Changi Airport today, views today’s launch as a significant milestone for TransNusa.

“Changi Airport is a world class airport with stringent requirements. The fact that TransNusa has managed to obtain all necessary approvals in such a short timeframe, reflects the importance and commitment we have towards safety, security, maintenance and aircraft performance measures,” Datuk Bernard added.

Today’s launch of the Jakarta – Singapore international route, comes at the back of TransNusa’s launch of its Jakarta – Guangzhou route on November 16th. TransNusa became the second Indonesian airline to receive approvals to start scheduled flight operations to Guangzhou, Guangdong, China.

Since April this year, TransNusa has introduced and launched scheduled flight operations in major capitals in South East Asia and Asia Pacific. The current destinations that TransNusa flies to are Singapore, Kuala Lumpur, Johor and Guangzhou. In the domestic front, the airline, which operates as a low-cost carrier, has scheduled flight to Bali and Jogjakarta.

On the airline’s future expansion, Datuk Bernard said “We have plans to further expand our international routes and we want to assist more passengers travel with ease to their destinations,” adding that he expects TransNusa to experience a significant growth in 2024.

On the commercial flights to Singapore, Datuk Bernard explained that TransNusa will be operating one round trip flight daily, with the hopes of increasing the frequency in the near future.

The TransNusa flight will depart Jakarta at 07.45am and arrive in Terminal 2, Singapore Changi Airport at 10.40am while flights from Singapore to Jakarta will depart at 11.40am and arrive in Jakarta at 12.25pm. Tickets for this route will be priced from SGD99.

Datuk Bernard had said previously that for its international routes, the services that TransNusa offers exceeded that of a low-cost airline.

“For our international flights, we not only provide premium services with competitive ticket prices in comparison to other low-cost airlines, but we have attractive new product bundles called SEAT, SEAT-PLUS and FLEXI-PRO.

GA-ASI Demonstrates Short Takeoff/Landing of UAS on UK Carrier

SAN DIEGO, CA, Nov 20, 2023 – (ACN Newswire) – General Atomics Aeronautical Systems, Inc. (GA-ASI) conducted a first-of-its-kind demonstration of its short takeoff and landing (STOL) capability on the HMS Prince of Wales, a Royal Navy aircraft carrier, using the Mojave Unmanned Aircraft System. The demonstration took place on November 15, 2023, when the Prince of Wales was underway off the East Coast of the U.S., and the Mojave was controlled by an aircrew within a control station onboard the ship. The demonstration included takeoff, circuits, and approaches and ended with a landing back onto the carrier.

Mojave is a short takeoff and landing UAS demonstrator originally developed to prove STOL operations at unprepared landing sites. While Mojave shares common systems and components with GA-ASI’s Gray Eagle model, a STOL wing set option is likewise being planned for the larger, more capable MQ-9B aircraft, which includes SkyGuardian(R), SeaGuardian(R), and the new Protector RG Mk 1 currently being delivered to the UK Royal Air Force. The MQ-9B version, called MQ-9B STOL, is being considered by the Royal Navy and other navies that operate aircraft from large flat-deck warships without catapults and arresting gear.

Royal Navy Director Develop, Rear Admiral James Parkin, whose team planned the trial, said: “The Mojave trial is a European first – the first time that a Remotely Piloted Air System of this size has operated to and from an aircraft carrier outside of the United States. The success of this trial heralds a new dawn in how we conduct maritime aviation and is another exciting step in the evolution of the Royal Navy’s Carrier Strike Group into a mixed crewed and uncrewed fighting force.”

Equipping UAS with STOL capability provides greater versatility and allows the aircraft to operate in areas previously deemed unsuitable for UAS operations, including landing onto and taking off from an aircraft carrier. MQ-9B STOL will be capable of carrying the same payloads and conducting the same missions as the SkyGuardian and SeaGuardian, including maritime surveillance, Anti-Submarine Warfare (ASW), Airborne Early Warning (AEW), and surface strike.

China-incorporated Fenglin Group, the administrative arm of the Shanghai Xuhui government in charge of Life Sciences, aims to establish an integrated ecosystem of local and international stakeholders in Shanghai’s Xuhui District to accelerate biopharmaceutical innovation and development.

1. Ordinary Resolution: Appointment of Foo Kon Tan LLP as the statutory auditor of the Company

2. Special Resolution: Change of name of the Company from “Kitchen Culture Holdings Ltd.” to

“SDAI Limited”

At the EGM, 99.997% of the approximately 184.59 million votes represented, were cast in favour of the two resolutions and the resolutions were duly passed.

The Company previously specialised in the sale and distribution of a wide range of premium imported kitchen systems, appliances, wardrobes systems, household furnishings and accessories from Europe and the USA. The Company worked closely with developers to market “higher-end” residential projects. Since 12 July 2021, the Company’s shares have been suspended due to a variety of issues including non-compliance and systemic internal failures.

Appointment of new Statutory Auditor, Foo Kon Tan LLP

The appointment of the new statutory auditor of the Company is a significant milestone accomplished by the present Board to rectify historical breaches of the Company, signifying a major step to work towards resumption of trading of the Company’s shares. Prior to this, the Company had failed to secure a statutory auditor when the previous auditor, Nexia TS Public Accounting Corporation (now known as CLA Global Public Accounting Corporation), did not seek re-appointment at the last Annual General Meeting of the Company held on 18 March 2022 for the financial year ended 30 June 2021. Consequently, the Company has failed to comply with the requisite listing rules of SGX and is in breach of statutory requirements under the Companies Act 1967 of Singapore for filing of statutory accounts and convening of shareholders’ meeting.

Change of the Company’s name to “SDAI Limited”

The change of the Company’s name from “Kitchen Culture Holdings Ltd.” to “SDAI Limited” will reflect the Company’s strategic direction and business activities going forward. This will enable the Company to create a new corporate identity for itself and develop a new positioning in the market, allowing the public and the Company’s business partners to better identify with the Company under this new name moving forward.

Going Forward

The Company will lodge the requisite statutory returns with the Accounting and Corporate Regulatory Authority of Singapore (“ACRA”) to effect the change of name of the Company from “Kitchen Culture Holdings Ltd.” to “SDAI Limited”. Upon the lodgement of the requisite statutory returns with ACRA, the Company shall adopt the name “SDAI Limited” as its new name and the name “SDAI Limited” shall replace all references to “Kitchen Culture Holdings Ltd.”.

The Company has also been working hard to accelerate the process of completing the Independent Special Audit being conducted by Deloitte & Touche Financial Advisory Services Pte. Ltd. by the end of the year, in compliance with statutory requirements, and is another key factor in the resumption of trading of the Company’s shares.
